This thesis addresses the question of what is the signaling pathway behind the metabotropic glutamate (mGlu) receptors 2 and 3 and how acute or prolonged exposure to the agonist LY379268 changes signaling transduction protein expression in brain regions related to reward, decision-making, and cognition. mGlu 2/3 receptor stimulation has been shown to be critical for regulation of neurotransmitter release and to have neuroprotective effects. Sprague Dawley rats were exposed acutely or chronically to 1.0 mg/kg/day or to 3.0 mg/kg/day doses of LY379268. The fixed brain slices were immunostained with CREB and ERK antibodies for the total and phosphorylated forms of the protein. This study found that acute exposure to 1.0 mg/kg/day of LY379268 increased total CREB protein expression while acute and chronic exposure to 3.0 mg/kg/day of LY379268 decreased total CREB protein expression. Likewise, acute and chronic exposure to 3.0 mg/kg/day of LY379268 decreased phosphorylated CREB to total CREB protein expression ratio. ERK total protein levels decreased after acute exposure to 1.0 mg/kg/day of LY379268.
