   <dim:field mdschema="dc" element="description" qualifier="abstract" lang="en_US">The synthesis of an azide-bearing &amp;lt;italic&amp;gt;N&amp;lt;/italic&amp;gt;-mustard cofactor, 8-azido-5&amp;amp;rsquo;-(diaminobutyric acid)-&amp;lt;italic&amp;gt;N&amp;lt;/italic&amp;gt;-iodoethyl-5&amp;amp;rsquo;-deoxyadenosine ammonium hydrochloride (&amp;lt;bold&amp;gt;2.1&amp;lt;/bold&amp;gt;), has been accomplished with a small amount of impurity in several steps from commercially available 2&amp;amp;rsquo;,3&amp;amp;rsquo;-isopropylidene adenosine (&amp;lt;bold&amp;gt;2.2&amp;lt;/bold&amp;gt;).  This cofactor was designed to efficiently mimic &amp;lt;italic&amp;gt;S&amp;lt;/italic&amp;gt;-adenosyl-L-methionine (SAM) by incorporating an &amp;lt;italic&amp;gt;N&amp;lt;/italic&amp;gt;-mustard and an amino acid moiety surrounding an adenosine core.  In addition, an azide functionality was introduced for post-alkylation modification.  The crucial factors that led to this success were (1) the choice of the proper alcohol protecting group and (2) the installation of the azide functionality at the &amp;lt;italic&amp;gt;C&amp;lt;/italic&amp;gt;8 position of the adenine base prior to the incorporation of &amp;lt;italic&amp;gt;N&amp;lt;/italic&amp;gt;-mustard and amino acid moieties.  Cofactor &amp;lt;bold&amp;gt;2.1&amp;lt;/bold&amp;gt; was found to be effectively transferred onto DNA by &amp;lt;italic&amp;gt;M.TaqI&amp;lt;/italic&amp;gt;, providing an azide-bearing modified DNA that will allow for subsequent chemoselective ligation chemistry that is hypothesized to facilitate the detection of biological methylation sites.</dim:field>
